Effects of Host Metabolism on Antibiotic Susceptibility
In order to understand if either infection or antibiotic treatment alters host metabolism, we did targeted, quantitative metabolomics on tissues from mice subjected to acute peritoneal infection and/or oral antibiotic administration. We found locally confined changes in host metabolism associated with infection, but globally distributed changes in metabolism associated with antibiotic treatment. Preliminary analyses of the metabolic changes quantified in the peritoneum suggest that bacterial infection remodels the local environment to help bacteria become more tolerant against antibiotics. This work is now being extended to further phenotype effects of host metabolism on bacterial pathogenicity and to identify candidate metabolic adjuvants that may be efficacious in host settings.
